The Onset of Action of a New Combination of Short and Long-Acting Methylprednisolone in Normal Volunteers

Thomas J. Vecchio M.D., F.A.C.P.1

1 Medical Research, Upjohn International, Inc., Kalamazoo, Michigan.

The eosinopenic and hyperglycemic activities of a new combination containing in each cc 20 mg of soluble methylprednisolone sodium succinate (MSS) and 40 mg of insoluble methylprednisolone acetate (MA) were compared with those of its individual components and the vehicle alone in normal prison volunteers. The onset of these activities was significant 2 hours after injection of the combination, whereas after MA alone significant eosinopenic activity was first observed in 8 hours, and significant hyperglycemic activity was not observed through 10 hours. Tolerance to the new combination was good.

Note:

I am grateful for invaluable assistance from the staff of the Upjohn Experimental Clinic at Jackson Prison and the Upjohn Clinical Research Laboratory in conducting this study and running the laboratory determinations, as well as for the statistical analysis performed by J. R. Schultz and J. R. Assenzo.
